Collection of saliva is by expectoration into metal-free tubes; collection devices such as the parotid cup or the Brock-Brotman collector may be used to avoid contamination especially by mercury from amalgam tooth fillings.

Intervening steps include: 1) sample collection, storage and preparation for analysis, 2) establishment and use of an adequate quality assurance control framework, 3) measurement of the element's concentration by some type of instrument using quantitation methods and 4) recording of the value obtained. In this section the methodologies for the quantitative analysis of metals in biological media are discussed.
